ഇംഗ്ലണ്ടിൽ നിന്ന് കുടിയേറിയ ജൂത മാതാപിതാക്കളായ ലീയുടെയും ജോസഫ് പ്ലോട്ട്കിന്റെയും മകനായി ന്യൂയോർക്ക് സിറ്റിയിലാണ് പ്ലോട്ട്കിൻ ജനിച്ച് വളർന്നത്. ന്യൂയോർക്ക് നഗരത്തിലെ ബ്രോങ്ക്സ് ഹൈ സ്കൂൾ ഓഫ് സയൻസിൽ പഠിച്ചു. ബ്രോങ്ക്സ് സയൻസിൽ പഠിക്കുമ്പോൾ പതിനഞ്ചാമത്തെ വയസ്സിൽ തന്റെ ഭാവി വിദ്യാഭ്യാസത്തെയും കരിയർ തിരഞ്ഞെടുപ്പുകളെയും വളരെയധികം സ്വാധീനിച്ച ഒരു ജോടി പുസ്തകങ്ങൾ സിൻക്ലെയർ ലെവിസിന്റെ ആരോസ്മിത്തും പോൾ ഡി ക്രൂയിഫിന്റെ മൈക്രോബ് ഹണ്ടേഴ്സും അദ്ദേഹം വായിച്ചു. ഒരു ഫിസിഷ്യനും ഗവേഷണ ശാസ്ത്രജ്ഞനുമായി ജീവിതം സമർപ്പിക്കാൻ തീരുമാനിച്ച പ്ലോട്ട്കിൻ 1948 ൽ ബ്രോങ്ക്സ് സയൻസിൽ നിന്ന് ബിരുദം നേടി. തുടർന്ന് 1952 ൽ ന്യൂയോർക്ക് യൂണിവേഴ്‌സിറ്റിയിൽ നിന്ന് ബിരുദം നേടി 1956 ൽ സുനി ഡൗൺസ്റ്റേറ്റ് മെഡിക്കൽ സെന്ററിൽ നിന്ന് എംഡി നേടി.
